چکیده

Among the various sensing principles proposed for bioaffinity studies, optical evanescent wave techniques have gained the lead in popularity. Next to evanescent ellipsometry [1] and the various optical waveguide platforms [2,3], surface plasmon resonance (SPR) spectroscopy [4–6], in particular, has found widespread applications and has demonstrated its potential for the sensitive detection of bioanalytes in numerous examples [7–9]. Since its introduction as a method for bioaffinity studies in 1983 by Liedberg et al. [10] and since the presentation of the first commercial instrument by Biacore in 1990 [11], the number of papers published has grown exponentially to currently more than 1500 contributions each year (Figure 9.1). This success story is largely based on the fact that SPR represents a label-free detection principle – the mere presence of the bound analyte slightly changes the optical architecture at the sensor surface which is probed by the surface plasmon mode propagating along this

Creating advanced multifunctional biosensors with surface enzymatic transformations.

This paper summarizes our recent work on the coupling of surface enzyme chemistry and bioaffinity interactions on biopolymer microarrays for the creation of multiplexed biosensors with enhanced selectivity and sensitivity. The surface sensitive techniques of surface plasmon resonance imaging (SPRI) and surface plasmon fluorescence spectroscopy (SPFS) are used to detect the surface enzymatic tra...

Plasmon-controlled fluorescence: A new detection technology.

Fluorescence is widely used in biological research. Future advances in biology and medicine often depend on the advances in the capabilities of fluorescence measurements. In this overview paper we describe how a combination of fluorescence, and plasmonics, and nanofabrication can fundamentally change and increase the capabilities of fluorescence technology. This change will be based on the use ...

A Microwell-Printing Fabrication Strategy for the On-Chip Templated Biosynthesis of Protein Microarrays for Surface Plasmon Resonance Imaging.

A two-step templated, ribosomal biosynthesis/printing method for the fabrication of protein microarrays for surface plasmon resonance imaging (SPRI) measurements is demonstrated. In the first step, a sixteen component microarray of proteins is created in microwells by cell free on chip protein synthesis; each microwell contains both an in vitro transcription and translation (IVTT) solution and ...

Advanced Methods for SPR Imaging Biosensing

Microarray biosensors have become an invaluable biotechnological tool for the rapid, multiplexed detection of surface bioaffinity interactions. For example, nucleic acid microarrays are currently being applied to the areas of genomics [1,2], genetic testing [3], gene expression [4,5], and single nucleotide polymorphism (SNP) genotyping [6,7]. Many researchers are also interested in developing p...

Plasmon light scattering in biology and medicine: new sensing approaches, visions and perspectives.

Surface plasmons are collective oscillations of free electrons at metallic surfaces. These oscillations can give rise to the intense colors of solutions of plasmon resonance nanoparticles and/or very intense scattering. While the use of plasmonic particle absorption based bioaffinity sensing is now widespread throughout biological research, the use of their scattering properties is relatively i...
